尿中美沙酮及代谢物的检测

通过考察尿中盐酸美沙酮及其代谢物的提取方法,建立其气相色谱-氮磷检测器(GC-NPD)和气相色谱-质谱(GC-MS)的检测方法。采用固相萃取法对尿中美沙酮原体及其主要代谢物进行提取,利用GC-MS对其进行检测并用GC-NPD 对美沙酮原体进行含量测定。线性范围为1mg/L~40mg/L(r=0.998,n=5),美沙酮的平均回率为79.3%,RSD=6.1%。本实验研究建立了尿中盐酸美沙酮及其代谢物的快速、简便、准确、灵敏的检测方法,可作为快速检验吸毒人员尿中是否含有美沙酮的一种有效的方法。 1 仪器与试药
美国VARIAN-3400型气相色谱仪;美国FINNIGAN-SSQ710型质谱仪;美国DEC-5000/125型数据处理系统;C18固相柱(天津开发区富集色谱技术发展公司)。 2 方法与结果 色谱条件:色谱柱:进样口温度:柱温:BP-1(30m×0.25mm)石英毛细管柱;280°C;200°C(1min)~280°C(20°C/min);检测器温度:290°C;载气:氮气10psi;氢气4.5ml/min;空气175ml/min。 3. 讨论
为配合禁毒工作,公安机关经常需要对吸毒嫌疑人员尿液进行检验,认定滥用药物种类。本文建立的固相萃取、GC/MS法,可作为快速检验吸毒人员尿中是否含有美沙酮的一种有效的方法。代谢物EDDP的检出可作为判定吸食美沙酮的佐证。

Detection of Methadone and Its Main Metabolite in Urine

To examine the optimal extractive conditions of methadone and its metabolite from urine and establish a detection method of methadone by GC-MS and GC-NPD. A solid-phase extraction and gas chromatography-mass spectrometry(SPE-GC-MS) procedure for the simultaneous detection of methadone and its main metabolite in urine has been developed. The linearity of the methadone solution from 1mg/L to 40mg/L is fine, and correlation coefficient is 0.998..The average recovery were 79.3%, and the Relative Standard Deviation were 6.1%..It is an rapid, simple, accurate, and sensitive method. It is also an effective method for rapid detection whether there is methadone in urine of the drug user.
